Peran Endonuclease-G sebagai Biomarker Penentu Apoptosis Sel Amnion pada Kehamilan dengan Ketuban Pecah Dini
Abstract: The aim of this
study was to identify whether there is an association between endonuclease-G
and the apoptosis of amniotic cells in pregnancies with premature rupture of
the membranes. Through research study on the amnion cell apoptosis mechanism in
pregnancies with premature rupture of the membranes, it is possible to know
whether endonuclease-G is the biomarker of amnion cells apoptosis involved.
Methods: Analytic observational methods with cross-sectional study have been
used in this study design. Sample tissues from the ruptured amniotic membrane
of 41 pregnant women with premature rupture of the membranes were taken and
processed to be analyzed further; TUNEL methods were used for obtaining the
apoptosis index analysis of amnion tissue; Western-Blotting methods were used
in both the analysis of endonuclease-G (30 kDa/full length) expression and also
the analysis of caspase-3 (35 kDa/full length) expression. Data, consisting age
of pregnancies, endonuclease-G, caspase-3 and apoptosis index were analyzed
using the chi- square test. The results of the study showed that apoptosis was
significantly associated with the high level detection of endonuclease-G(p =
0.000) but was not associated with caspase-3 significantly (p = 0.316). It was
also found that the apoptosis in premature rupture of the membranes in
pregnancy is not dependent to the age of pregnancies. Therefore, there is an
indication that the occurrence of ptosis is dominant through the
caspase-independent pathway (endonuclease-G).Hence it can be concluded from the
results of this study that high level detection of endonuclease-G is a
biomarker of apoptosis of amnion cells in pregnancies with premature rupture of
the membranes.
