Perbandingan Aktivitas Enzim MMP-9, TIMP-1, dan Ekspresi Kolagen-4 pada Model SOPK Dibandingkan dengan Siklus Estrus Normal
Abstract: This study was aimed
to explain pathogenesis of occurring ovarian cyst and ovulation disturbance in
PCOS case. This study is an experimental study using Rattus novergicus as PCOS
model, compared to control study in every estrus cycle phase of Rattus
novergicus, ie, proestrous, estrous, metestrous and disestrous phase. Each
study group and control group consisted of 5 rats so that there were a total of
25 Rattus novergicus. PCOS model group was injected with testosterone
propionate with dose 1mg/ 100 BW, whereas control group with propylene glycol
injection. The examination of MMP-9, TIMP-1 activity by ELISA activity and
Collagen-4 used immunohistochemistry staining. The statistic analysis used
ANOVA test. The average of MMP-9 activity of PCOS model (P): 235.17 (SD: 5.47),
Proestrous Group (PG): 257.40 (SD: 0.25), Estrous Group (EG): 262.80 (SD:
2.34), Metestrous Group (MG):254.43 (SD: 6.21), Diestrous Group (DG):240.69
(SD: 4.16). With ANOVA test, PCOS model only indicated a significant difference
with Estrus Group: (p = 0.001). The average of TIMP-1 activity of P: 57.15(SD:
2.46), PG: 51.63(SD: 3.81), EG: 16.30 (SD: 2.33), MG: 12.84 (SD: 0.42 ), DG:
28.95 (SD: 3.07). With ANOVA test, PCOS model indicated a significant
difference with Estrus, Metestrus and Diestrus (p = 0.000).The average of
Collagen – 4 expression P: 41.60 (SD: 3.36 ), PG: 22.40 (SD: 3.51), EG: 21.40
(SD: 1.52), MG: 23.80 (SD: 1.92), DG: 23.80 (SD: 1.79) with ANOVA test
indicated a significant difference (p = 0.00). The decrease of MMP-9 activity
and the increase of TIMP-1 activity occuring in PCOS model have resulted in the
increase of Collagen 4 expression and finally caused the thickening fibrous
follicle wall consequently, the failure of follicle enlargement may
mechanically occur.
