Analisis α-Mangostin Dalam Minuman Herbal Kulit Buah Manggis (Garcinia mangostana L.) dengan Metode Kromatografi Lapis Tipis-Densitometri
ABSTRACT: These days,
mangosteen juice is becoming a popular “herbal drink.” Some marketers claim
that mangosteen juice can treat diarrhea, menstrual problems, urinary tract
infections, tuberculosis, and a variety of other conditions. However, there is
no reliable scientific evidence to support these claims. This study aims to determination of
α-mangostin in the herbal drinks of mangosteen rind on the market by thin layer
chromatography-densitometric method. The analysis was performed with TLC
Scanner using Camag TLC Scanner 4 with Wincats software, the maximum wavelength
of α-mangostin was 316 nm, viewed using 254 nm and 366 nm UV Camag lamps, Camag
Nanomat 4, Camag capillary pipe size 2 μL, Camag Twin Chamber, silica gel plate
60 F254 (Merck) size 20 x 20 cm. The mobile phase used was chloroform and ethyl
acetate (9: 1). After being seen under a 254 nm UV lamp, the α-mangostin
standard, samples B and C have the same Rf value of 0.48. The concentration
series used 50, 100, 150, 300 and 400 ppm yield regression equation y =
1326,1658 + 19,7647x with correlation coefficient (r) = 0,99415. This
correlation coefficient shows a linear result. Limit detection (LOD) and
quantification limit (LOQ) obtained were 54.38 μg/mL and 181.28 μg/mL. The percentage
of recovery obtained from the analysis of 90.04%, this validation method gives
accurate and precise results (intraday precision value RSD / Relative Standard
Deviation was ≤ 1.28%, while the interday precision test was ≤ 2.39%). The
average result obtained for samples B and C were 0.040% w/v and 0.118% w/v,
respectively. This research is cheap, accurate, precise, linear and fast. The
method of TLC-densitometry meets the validation criteria for the quantitative
determination of α-mangostin in the herbal drinks of mangosteen rind.
